Temperature effects on lethal mutation rates of Habrobracon oocytes x-irradiated in first meiotic metaphase.

چکیده
BRIEF note was published in 1940 (WHITING) concerning the influence of A temperature on X-ray sensitivity of different meiotic stages in the Habrobracon oocyte. Unmated females were kept at O"C, 13"C, 25"C, and 35OC for one hour before and one hour after irradiation. During a one half minute of exposure to 212r, they were at room temperature. For oocytes in first meiotic metaphase (metaphase I), mortality was lowest at O"C, highest at 25°C and intermediate at 35°C. The consistent and extensive radiation data on metaphase I obtained since 194.0 made it seem valuable to redesign and to repeat the experiment. When Habrobracon females are X-irradiated, responses of their oocytes in prophase I can be distinguished from those in metaphase I (WHITING 1945). The latter were used in the experiments reported in this paper. LD 50 for the stage is about 400r. The nature of the dose-action curve as well as the absence of any significant differences in response of this curve to different experimental conditions (for example, 124 kv, 50 mev by HEIDENTHAL, C ARK and GOWEN 1955; lOOr/min X-rays, 2OOrJsec cathode rays by HEIDENTHAL 1960; 6.6r/min, SOOr/ min by LACHANCE 1960; and fractionation by WHITING 1945) furnish evidence for one-hit X-ray-induced changes. Tetrad conditions at the time of irradiation and cytological observations on succeeding stages also make the one-hit explanation a plausible one. Viable eggs laid by unmated females develop into haploid males. Those in which dominant or recessive lethal mutations acting on the embryo are induced (as happens in the majority of eggs) fail to hatch.

INVESTIGATIONS to be reported in this paper have been concerned chiefly whh X-ray-induced embryonic lethal mutations in Habrobracon. Some of the data and one of the methods have already been given (HEIDENTHAL 1952, 1953; HEIDENTHAL, C ARK and GOWEN 1955). The purpose of the author's work, as well as that of others, has been in part to study the frequency of different kinds of lethal-bearing gen...

HE technique for classifying oocytes of irradiated females of the parasitic Twasp, Habrobracon, into those exposed in first meiotic prophase (prophase I ) and those exposed in first meiotic metaphase (metaphase I) was described by the author in 1945 (1945a). Methods for greater accuracy in estimating lethal rates were described by ATWOOD, VON BORSTEL and WHITING 1956. The striking difference in...

HITING (1945), working on the parasitic wasp Habrobracon, developed a timing method that distinguished oocytes irradiated in the first meiotic metaphase (metaphase I) from those irradiated in the first meiotic prophase (prophase I) . She used embryo lethality of X-irradiated eggs from diploid virgins, which measured mostly dominant lethality, and found that metaphase I oocytes are approximately...

IFFERENCES in sensitivity of oocytes to X-irradiation depending upon stage in meiosis when treatment occurred, have been shown for a number of organisms including Habrobracon (WHITING l938,1941,1945a, 1945b; ATWOOD, VON BORSTEL and WHITING 1956). Studies have also shown that death of Habrobracon embryos following irradiation of oocytes in late metaphase and in late prophase (diplotene) was attr...

HEN Habrobracon eggs are X-rayed at metaphase I and subsequently fertiw l i z e d , the decreased hatchability is attributed to dominant lethal mutation, in the same sense in which the term is used when sperm are irradiated. This is justified by WHITING'S demonstration (1946, 1948, 1949, 1950, 1955) that cytoplasmic injury is not lethal a t the doses employed. In unfertilized eggs, which develo...
